Gran èxit de l'Open Day 2009Society meets Science

Society meets Science
The last PRBB Open Day on Saturday, October 3, dedicated to Darwin, was
a great success. This initiative was proposed two years ago by the Science meets Society (SmS) group, and it was only possible thanks to the support from the PRBB research centers and sponsors, but mainly to the dedication of many volunteers. An informed and welleducated society forms the basis of a true democratic system, and the achievement of a widespread scientific culture opens doors for more comprehension. Starting at child hood, citizens must be motivated to think, to be curious, to participate and to ask “why”. Therefore, initiatives like the Open Day can only bring benefits. The Open Day also crossed language barriers, judging by the high number of nonnative speaking volunteers. In part, it is the multiculturalism and diversity that makes a place like PRBB so fascinating. This example could perhaps be extended to other sectors of society.

Gràcies a tots!
Great succes of the Open Day 2009
Last October 3 the weather and the music were just right for the PRBB Open
Day. The inner square exposed an impressive decoration that simulated a sailing boat where the group PeRaBaBaus presented twice the theatre play Evolution, what a revolution! The bar, the science stands and the rooms for multimedia and video games were open all day. At the terrace of the first floor the exposition about Darwin was magnificent and at the Charles Darwin Square children and adults did all kinds of experiments. Inside the building the visiting groups filled the corridors and laboratories, and in the afternoon a scientific coffee
gathered together an interested and lively audience that did not seem to want to leave. At the end a Catalan drum show by the group Karabassà closed the event. The statistics of the attendance went beyond the most optimistic predictions. Just for the guided visits (only acti vity that required an inscription) 1,647 vi sitors were counted and total attendance is estimated to be about 3,500 people. Concerning the public opi ni on, in the collected surveys we got a very good grade for both the organization and the contents and they encoura ge us to repeat next year. All this has only been possible thanks to the 156 volunteers that worked bravely. Thank you all!

for treating this type of lymphoma. This group of experts, in collaboration with the Ramon y Cajal Hospital in Madrid and the Barcelona ICO, have shown that adding an antibody that recognizes B cells to conventional chemotherapy represents signif icant advantages, because defensive cells detect this antibody and attack the cells that are labelled with it
“Sequencing the genomes will be the easy part”
What is the aim of the International Can-cer Genome Consortium?
More than the actual number of sequenced genomes, the aim is to identify the molecular mechanisms
involved in different types of cancer. Due to the heterogeneity found within each cancer, it is necessary to sequence about 500 samples of each type in order to have a representative vision. Another important objective is to create a cooperative work environment to avoid the need of reinventing everything for each type of tumour. For this reason the protocols are shared, and quality control and data analysis standards have been developed.
How do you discover the mutations? We sequence two samples of each subject, one from healthy tissue and one from the tumour. Each sample is sequenced up to 50 times, to be sure that we have the whole genome well covered. When the sequence has been assembled, we compare the healthy and the tumoral sequences.
We expect to find about 30,000 diffe rences, which can vary from changes in a single nucleotide to structural changes, such as insertions, deletions or repetitions of DNA. But not all of these changes will be involved in causing the cancer. Some of these mutations are drivers, which are the ones who cause the cancer, while others are passengers, because they accompany the cancer, and exist as a consequence of it. In order to distinguish them, and to know which are the specific mechanisms by which each mutation is involved in the tumour, we will need many more functional studies.
So, the project goes further than se-quencing itself? Yes, to have the sequence of the genome will be, in fact, the easiest task! Afterwards come all the analyses: not only the genomic comparisons, but also the studies of the transcriptome (which genes are transcribed and at what levels, which non coding RNAs are expressed or what variants of splicing take place) and the epigenome (DNA methylation studies).
How many people form the consortium? In the last meeting there were about 150 people, but a lot more are working on it. Every group who wants to be part of the consortium proposes a type of cancer and has to contribute with 20 million dollars. The Spanish consortium studies the chronic lymphocytic leukaemia and is formed by eight research centres and universities. At the PRBB level, Roderic Guigó (CRG), Núria LópezBigas (GRIB: IMIMUPF) and myself are part of the consortium.
What is your role in the project? Now we are at a pilot stage, until March 2010. We are sequencing the RNA and studying the structural variations of about 50 samples. Out of these, three are being totally sequenced, but they are doing it at the Sanger Institute, in the UK, because we don’thave enough capacity here. Next year we will have the first sequencing centre in Spain, located at the Parc Científic and then we will do the global sequencing from here. On the other hand, Núria LópezBigas is part of the analysis coordination group and has developed a bioinformatic tool that will be very useful to integrate all the information generated

“Radiation is neither good nor bad, it depends on the use we make of it”
Still located at the Hospital de l’Esperança, the IOR (Institute of Radiotherapy and Oncology) is a medi
cal service of the IMAS that will move to the Hospital del Mar towards 2011, with the hospital enlargement. There are currently seven doctors specialized in radiotherapy, three physicians specialized in medical radiophysics, and 22 more people amongst technicians, nurses and administrative staff. They all divide their time between research, clinical practice and teaching. At the IOR they are leading several clinical essays, one of them a study on radiotherapy dose escalation. “Lung cancer is one of the most difficult ones to treat. The solution is to increase the dose, but this would have negative consequences. We know that the tumour zones that receive less oxygen (hypoxic) are those more resistant to radiotherapy, since cells in those areas don’t divide. So the idea is to label the most hypoxic areas using PET and to increase the dose only in those zones”, explains Manel Algara, director of the service. In order to increase the dose without increasing the toxicity, technical advances are needed constantly. “We have been using the new technologies of IntensityModulated Radiation Therapy (IMRT) for a year.
These technologies allow dose escalation, but they are very complex, not all the machines are prepared to use them, and the treatment is more complicated and longer”, emphasizes Martí Lacruz, head of physics of the service. Another essay performed at the IOR, this one in collaboration with the anatomical pathology service of Hospital del Mar, tries to foresee which patients will present a high toxicity if treated of prostate cancer, in order to decide whether to do the treatment at all. For that, they are doing a study about radioinduced apoptosis (cell death) in lymphocytes of the patients. Even though research is an important part of the job of the IOR staff, most of their time is dedicated to clinical practice. There are two types of radiotherapy, external and internal. External radiotherapy makes it possible to irradiate big volumes, for example an entire breast. “Treatments are done with linear particles accelerators that emit a ray that penetrates the patient”, says Algara. The internal radiotherapy, or brachytherapy, is used for smaller tumours or those that are focalized and accessible, and it takes advantage of natural holes in the body to introduce the radioactive isotope inside the body. “Gynaecological brachytherapy is done with a higher rate of dose (HRD), the most modern that exists", explains Lacruz. In both external and internal radiotherapy, the radiation machines are accompanied by software. “First we do a scanner to localize what we want to treat. We also use PETTAC, in collaboration with CRCCIM (IAT) at the PRBB, in which the images from the scanner and PET are fused”, details Lacruz. “Then we use a virtual simulation system to better define the volumes that we want to treat, and finally the physicians calculate the distribution of the optimum dose in a 3D dosimetric planning system”. Six of the members of the IOR are also associated teachers at the CEXSUPF, where they teach physics for the biology and for the medicine students. Talking about their connection to the PRBB, Manel Algara says: “Once we are physically at the Hospital del Mar we would like to offer all our potential to irradiate animals and collaborate with research groups from the PRBB”

They confirmed that the cannabinoid system is actively involved during the inflammatory phase of the disease and the presence and distribution of its elements changes according to the degree of inflammatory activity of the tissue. Furthermore the results suggest some antiinflammatory properties of this system. Thus, the analysis of the endocannabinoid system could be useful i n the d iagnos is and as sessment of the severity of this process, and the use of cannabinoid drugs might be an alternative therapy for ulcerative colitis.

llarg i costós.
Computer-assisted drug design
The development of a new drug with the characteristics of efficacy and safety required today is a long and
costly process. Typically, the process can last for more than 12 years and require highly risky investments superior to one billion euros. In this scenario it is not surprising that the pharmaceutical companies are open to incorporate novel technological advances,
with the hope of making the process faster and more efficient. The term “computer assisted drug design” (CADD) encompasses a wide range of technologies which have in common the application of computatio nal methods at diverse points of the drug development pipeline. Focusing on the preclinical stages, CADD techniques are typically applied in the search of drug candidates with a certain profile of biological properties. When the structure of a biological receptor is known, direct methodologies can exploit this information to search for molecules that fit well the receptor binding site. This can be done by different methods, for example assembling fragments which complement the shape and physicochemical properties of the receptor, or searching for a suitable compound in databases storing millions of structures. When the structure of the receptor is not available, but a few lowactive compounds are known, it is still possible to apply methods based on molecular similarity to obtain better compounds. In practice, the methods are more complex because several biological properties must be considered and optimized simultaneously. Indeed, modern approaches to drug design do not consider a single target but the interaction of the candidate with many receptors, some asso ciated with desirable properties and others associated with undesirable side effects
